We extracted the DNA melting curve and its derivative from each PCR cycle during the transition from the elongation to the denaturation temperature with a set heating rate of 0.8 K· s−1and then used the data to construct individual PCR amplification curves for each gene to determine the initial concentration of DNA in the sample. In the framework of the Poland Scheraga model of DNA denaturation, we derive a recursion relation for the partition function of double stranded DNA, allowing for mismatches between the two strands. Heating DNA in the presence of a fluorescent DNA‐binding dye generates melting curves. The resultant decrease in fluorescence arises from background loss of fluorescence as the temperature increases and from DNA denaturation and consequent release of the DNA‐binding dye. Then, there is a dramatic increase in the second part of the curve (60oC-80oC). It means that many DNA molecules undergo thermal denaturation.
